Src family of tyrosine kinases are directly involved in memory. Inhibiting these kinases with PP2 in the hippocampus blocked short-term and long-term memory formation and recall in rats.

Area of Science:

  • Neuroscience
  • Molecular Biology
  • Pharmacology

Background:

  • The Src family of tyrosine kinases are implicated in various cellular processes.
  • Their specific role in memory formation and recall remains to be elucidated.

Purpose of the Study:

  • To investigate the direct involvement of Src family tyrosine kinases in the hippocampus during memory formation and recall.

Main Methods:

  • Rats were trained in a one-trial inhibitory avoidance task.
  • PP2, a specific Src kinase inhibitor, or its inactive analog PP3 were infused into the CA1 region of the dorsal hippocampus at various time points relative to training and testing.
  • Memory formation (STM and LTM) and recall were assessed at different time points post-training and pre-testing.

Related Experiment Videos

Main Results:

  • Infusion of PP2 immediately or 30 min after training blocked both short-term memory (STM) and long-term memory (LTM) formation.
  • PP2 administration at later time points (60 min post-training) did not affect STM or LTM formation.
  • PP2 infused 15 min before a long-term memory expression test blocked memory recall.

Conclusions:

  • Activity of Src family tyrosine kinases in the CA1 region of the dorsal hippocampus is essential for the formation and retrieval of one-trial inhibitory avoidance memory.
  • These findings highlight a critical role for Src kinases in hippocampal memory processes.
