Summary
Aldosterone-induced serum and glucocorticoid-regulated kinase-1 (SGK1) may coordinate the cell-surface expression of the Na,K-ATPase pump and epithelial sodium channel (ENaC). This study suggests SGK1
Area of Science:
- Nephrology
- Molecular Biology
- Cell Biology
Background:
- Aldosterone regulates sodium balance by increasing cell-surface expression of epithelial sodium channels (ENaC) and Na,K-ATPase pumps in the distal nephron.
- The role of serum and glucocorticoid-regulated kinase-1 (SGK1) in the coordinated regulation of these transporters is not fully understood.
Purpose of the Study:
- To investigate whether SGK1 mediates the aldosterone-induced regulation of Na,K-ATPase subcellular localization.
- To determine if SGK1 specifically affects the cell-surface expression of Na,K-ATPase.
Main Methods:
- Co-expression of rat Na,K-ATPase (alpha 1 and beta 1 subunits) and Xenopus SGK1 in Xenopus oocytes.
- Measurement of Na(+) pump current to assess Na,K-ATPase function.
- Western blotting of surface-biotinylated proteins to quantify cell-surface expression.
- Assessment of other transporters (LAT1-4F2hc, NaPi-IIa) for SGK1 specificity.
Main Results:
- Wild-type SGK1 increased the function and cell-surface expression of exogenous Na,K-ATPase in Xenopus oocytes.
- SGK1 did not affect the surface expression of LAT1-4F2hc or NaPi-IIa, indicating specificity.
- SGK1 increased the total pool of Na,K-ATPase, an effect also observed with ENaC co-expression.
- SGK1 enhanced Na,K-ATPase surface expression even when the total pool was increased by ENaC.
Conclusions:
- SGK1 can increase the functional cell-surface expression of Na,K-ATPase.
- These findings suggest SGK1 may play a role in the coordinated regulation of Na,K-ATPase and ENaC in the distal nephron.
- SGK1's effect on Na,K-ATPase surface expression is specific and not a general effect on all transporters.
