Background:

  • Endothelin-1 (ET-1) and endothelin-3 (ET-3) affect hepatic blood flow.
  • Previous studies indicated ET-3 causes greater reductions in hepatic blood flow than ET-1.
  • The role of the arachidonic acid cascade, particularly thromboxane A2 (TXA2), in mediating these effects is not fully understood.

Purpose of the Study:

  • To investigate the involvement of the arachidonic acid cascade, specifically TXA2, in the changes in hepatic blood flow induced by ET-1 and ET-3.
  • To differentiate the mechanisms by which ET-1 and ET-3 alter hepatic microcirculation.

Main Methods:

  • Administered indomethacin (INDO), a prostaglandin (PG) biosynthesis inhibitor, prior to ET-1 and ET-3.
  • Administered OKY-046, a selective TXA2 synthesis inhibitor, prior to ET-1 and ET-3.
