Summary

CpcE and CpcF proteins from Synechococcus sp. PCC 7002 catalyze phycocyanobilin attachment to the alpha subunit of apophycocyanin. These proteins are essential for correct bilin attachment in phycobiliproteins.

Area of Science:

  • Biochemistry
  • Photosynthesis
  • Cyanobacterial research

Background:

  • Phycobiliproteins are light-harvesting proteins in photosynthesis with covalently attached bilin chromophores.
  • In Synechococcus sp. PCC 7002, phycocyanobilin is attached to cysteine residues via thioether bonds at eight sites.
  • The cpcE and cpcF genes are involved in bilin attachment, with inactivation affecting the alpha subunit of phycocyanin.

Purpose of the Study:

  • To investigate the function of CpcE and CpcF proteins in bilin attachment.
  • To characterize the in vitro activity of CpcE and CpcF in catalyzing bilin conjugation.
  • To determine the specificity of CpcE and CpcF for bilin attachment sites.

Main Methods:

  • Overproduction of CpcE and CpcF proteins in Escherichia coli.

Related Experiment Videos

  • In vitro assays to study the forward and reverse reactions of bilin attachment.
  • Site-directed analysis of bilin conjugation to the alpha subunit of apophycocyanin.
  • Main Results:

    • CpcE and CpcF proteins were successfully overproduced in E. coli.
    • In vitro, CpcE and CpcF catalyze the attachment of phycocyanobilin to the alpha subunit of apophycocyanin at alpha-Cys-84.
    • Both proteins are required for forward and reverse bilin transfer reactions, showing specificity for the alpha-Cys-84 position.

    Conclusions:

    • CpcE and CpcF are essential for the correct and specific attachment of phycocyanobilin to the alpha subunit of phycocyanin.
    • Phycocyanobilin serves as the immediate precursor for protein-bound bilin.
    • The study elucidates the enzymatic mechanism of bilin conjugation in cyanobacteria.
