Summary

Human adenoviruses (Ad) use clathrin-mediated endocytosis for entry into respiratory cells. Macropinocytosis also plays a role, especially when targeting other cell types, aiding viral infection.

Area of Science:

  • Cell biology
  • Virology
  • Molecular biology

Background:

  • Pathogen entry into host cells utilizes diverse mechanisms like endocytosis.
  • Human adenoviruses (Ad) infect respiratory epithelial cells via specific receptors.

Purpose of the Study:

  • To investigate the uptake mechanisms of human adenovirus (Ad) types 2 and 5.
  • To integrate Ad uptake into the broader context of cellular endocytosis.

Main Methods:

  • Survey of Ad uptake pathways.
  • Analysis of clathrin-mediated endocytosis and macropinocytosis in Ad infection.
  • Investigation of Ad interaction with receptors (CAR, integrins, Fcgamma receptor CD64).

Main Results:

  • Ad2 and Ad5 primarily use clathrin-mediated endocytosis involving dynamin and AP2.
  • Macropinocytosis is induced simultaneously and is crucial for Ad entry into hematopoietic cells via CD64.
  • Adenovirus infection modulates endocytic trafficking and disrupts cellular compartments.

Conclusions:

  • Adenovirus entry pathways are cell-type specific and can be modulated.
  • Understanding these pathways allows for targeted delivery of Ad vectors.
