Summary

Immature swine show greater tracheal smooth muscle (TSM) contraction due to less acetylcholinesterase (AChase) activity. Allergic dogs also exhibit increased TSM contraction, which is normalized by inhibiting AChase.

Background:

Purpose of the Study:

  • To investigate the developmental changes in TSM contraction and AChase activity in swine.
  • To examine the role of AChase in allergic bronchospasm in dogs.
  • To compare TSM responsiveness to ACh in immature versus mature swine and in allergic versus non-allergic dogs.

Main Methods:

  • Isometric tethering of TSM strips at optimal length.
  • Measurement of TSM contraction as a percentage of maximum KCl response (% KCl).

    • Allergic bronchospasm in dogs involves augmented TSM responsiveness to ACh.
    • AChase plays a significant role in modulating TSM contraction in both developmental and allergic contexts.
