Insights

Photodynamic therapy (PDT) significantly increases Mac-1 expression and leukotriene B(4) generation in polymorphonuclear leukocytes (PMNs). These effects suggest a role for PDT-induced PMN activity in oral cancer treatment.

Area of Science:

  • Biomedical Sciences
  • Oncology
  • Immunology

Background:

  • Photodynamic therapy (PDT) is an emerging cancer treatment modality.
  • Polymorphonuclear leukocytes (PMNs) play a crucial role in the immune response.
  • Mac-1 and leukotriene B(4) (LTB(4)) are key mediators in inflammatory processes.

Purpose of the Study:

  • To investigate the effect of N-aspartylchlorin e6 (NPe6)-mediated PDT on Mac-1 expression and LTB(4) generation by PMNs.
  • To explore the potential role of PDT-induced PMN activation in oral cancer therapy.

Main Methods:

  • PMNs were exposed to NPe6 and subsequently treated with PDT (10 J/cm(2), 664 nm).
  • Mac-1 expression was quantified using flow cytometry.
  • LTB(4) generation was measured using enzyme-linked immunosorbent assay (ELISA).
  • The effect of 5-lipoxygenase inhibitor (AA861) on Mac-1 expression and LTB(4) generation was evaluated.

Main Results:

  • PDT significantly increased Mac-1 expression on PMNs by approximately 3-fold compared to NPe6 exposure alone.
  • The increase in Mac-1 expression was not affected by the 5-lipoxygenase inhibitor AA861.
  • LTB(4) generation was enhanced by NPe6 incubation and further promoted by PDT.
  • PDT-induced LTB(4) generation was significantly inhibited by AA861.

Conclusions:

  • PDT effectively induces Mac-1 expression on PMNs.
  • PDT enhances LTB(4) generation by PMNs, an effect mediated by 5-lipoxygenase.
  • PDT-induced Mac-1 expression and LTB(4) generation in PMNs may contribute to the cytocidal activity observed in oral cancer treatment.
