Insights

Granulocyte-colony stimulating factor (G-CSF) protects microvascular endothelial cells (mECs) from LPS-induced apoptosis and reduces inflammatory IL-8 secretion. This suggests G-CSF

Area of Science:

  • Endothelial Cell Biology
  • Immunology
  • Hematology

Background:

  • Microvascular endothelial cells (mECs) are found in increased numbers in severe sepsis and hemophagocytic syndromes.
  • These circulating mECs are viable and can be used to establish cell lines.
  • Antigenic profiling of these mECs shows expression of CD36, factor VIIIa, CD95-ligand, CD44, and CD146.

Purpose of the Study:

  • To investigate the antioxidative and protective effects of the hematopoietic growth factor G-CSF on microvascular endothelial cells (mECs).
  • To determine the impact of G-CSF on lipopolysaccharide (LPS)-induced inflammatory responses and apoptosis in mECs.

Main Methods:

  • Microvascular endothelial cells (mECs) were stimulated with LPS (Escherichia coli 0111:B4) for 24 and 48 hours, with and without preincubation with G-CSF.
  • Interleukin-8 (IL-8) secretion was measured by ELISA.
  • Hemoxygenase-1 (HO-1) expression and caspase 3 activation were assessed by immunohistochemistry and flow cytometry.

Main Results:

  • LPS stimulation upregulated IL-8 secretion in mECs by 2-10 fold.
  • G-CSF preincubation significantly downregulated LPS-induced IL-8 secretion (20-50%) but did not affect IL-6 production.
  • LPS stimulation led to apoptosis and caspase 3 activation in mECs after 48 hours, which was markedly reduced by G-CSF preincubation.

Conclusions:

  • G-CSF exhibits a protective effect against LPS-induced inflammatory responses and apoptosis in microvascular endothelial cells.
  • G-CSF downregulates IL-8 secretion and caspase 3 activation in mECs stimulated with LPS.
  • These findings suggest a potential therapeutic role for G-CSF in conditions involving endothelial cell damage and inflammation, such as SIRS.

Calmodulin-dependent Signaling01:16

Calmodulin-dependent Signaling

Calmodulin (CaM) is a calcium-binding protein in eukaryotes that controls various calcium-regulated cellular processes. It has four calcium-binding sites that bind calcium to form the calcium-calmodulin ( Ca2+-CaM) complex. GPCR stimulation increases the calcium levels in the cells that bind to CaM and induces a conformational change.
The Ca2+-CaM complex does not have enzymatic activity by itself. Instead, the complex binds downstream target proteins, including membrane proteins or enzymes,...
The Extrinsic Apoptotic Pathway01:17

The Extrinsic Apoptotic Pathway

The extrinsic apoptotic pathway is initiated when extracellular death-inducing signals, such as specific cytokines, activate the death receptors expressed on the cell surface. The immune cells involved in this pathway are natural killer cells (NK cells) and cytotoxic T-lymphocytes. NK cells are critical in innate immune response, while cytotoxic T-lymphocytes are associated with adaptive immune response. These cells recognize specific receptors expressed on the altered cells and activate...
Regulation of Angiogenesis and Blood Supply01:24

Regulation of Angiogenesis and Blood Supply

Rapidly dividing tumors, embryos, and wounded tissues require more oxygen than usual, lowering the oxygen concentration in the blood. At low oxygen or hypoxic conditions, an oxygen-sensitive transcription factor called the hypoxia-inducible factor 1 or HIF1 is activated. HIF1 is a dimeric protein of alpha (ɑ) and beta (β) subunits.  Under optimal oxygen conditions, HIF1β is present in the nucleus while HIF1ɑ remains in the cytosol. HIF1ɑ is hydroxylated by prolyl hydroxylase and factor...
Regulation of Hematopoietic Stem Cells01:01

Regulation of Hematopoietic Stem Cells

All blood and immune cells are produced from the multipotent hematopoietic stem cells (HSCs) by the process of hematopoiesis. However, they all have a limited life span. In addition, many are depleted in immune surveillance or combatting an injury or infection. This makes blood one of the most regenerative tissues. Hematopoiesis helps replenish these blood and immune cells, restoring the body's normal functioning. However, overproduction of blood and immune cells can make them cancerous or...
Feedback Regulation of Calcium Concentration01:27

Feedback Regulation of Calcium Concentration

Calcium is an essential signaling molecule required for various cellular functions. Calcium pumps and ion channels on cell and organellar membranes, such as those on the endoplasmic reticulum (ER), regulate calcium concentrations inside the cell. They remain closed, keeping the cytosolic calcium levels low at a resting state.
Various transmembrane receptors, such as G protein-coupled receptors (GPCRs), elicit a response to extracellular signals by increasing cytosolic calcium. Activated GPCRs...
