Background:

Tityus serrulatus scorpion venom contains numerous toxic components.
Previous studies have identified various toxins, but a comprehensive understanding of their purification and characterization remains incomplete.

Purpose of the Study:

To fractionate Tityus serrulatus scorpion venom.
To identify and characterize the major toxic components, particularly the most potent toxin.
To differentiate between various protein fractions and their toxicological significance.

Main Methods:

Gel filtration chromatography was used to obtain two heterogeneous toxic fractions (T1 and T2).
Ion-exchange chromatography was employed to further fractionate the T2 fraction.
High-performance liquid chromatography (HPLC) and protein purification techniques were utilized.

Main Results:

Gel filtration yielded two toxic fractions, T1 and T2.
Ion-exchange chromatography of T2 revealed that the fraction originally named 'tityustoxin' was a mixture of proteins, including TsTX.
A highly purified protein, gamma-toxin, was isolated and identified as the major and potentially most potent toxin, distinct from TsTX.
Gamma-toxin was found to be present in both T1 and T2 fractions.

Conclusions:

The fractionation of Tityus serrulatus venom has led to the isolation and partial characterization of its major toxic components.
Gamma-toxin represents a significant and potent toxin within the venom, distinct from previously identified toxins like TsTX.
Further research into the specific mechanisms of action and structure of gamma-toxin is warranted.
