Pseudoxanthoma elasticum (PXE) typically follows autosomal recessive inheritance. While autosomal dominant inheritance is possible, this study indicates it is much rarer than previously believed, even in families with suggestive histories.

Area of Science:

  • Genetics
  • Dermatology
  • Ophthalmology

Background:

  • Pseudoxanthoma elasticum (PXE) is a genetic disorder affecting elastic fibers.
  • PXE is primarily caused by mutations in the ABCC6 gene.
  • While autosomal recessive inheritance is common, autosomal dominant (AD) PXE has been reported.

Observation:

  • This study reviewed literature and analyzed clinical/DNA data from 59 PXE probands and families.
  • Only three families with definite AD PXE across two generations were found in the literature.
  • Three putative AD PXE families were studied, with only one showing definite PXE in two generations and ruling out pseudodominance.

Findings:

  • Autosomal dominant inheritance in PXE is significantly rarer than previously assumed.
  • Most familial cases exhibit autosomal recessive inheritance patterns.

Related Experiment Videos

  • Phenotypic expression in heterozygous carriers of autosomal recessive PXE may mimic AD inheritance.
  • Implications:

    • Re-evaluation of suspected autosomal dominant PXE pedigrees is warranted.
    • Understanding inheritance patterns is crucial for genetic counseling and patient management.
    • Further research may clarify the role of ABCC6 variants in diverse inheritance patterns.
