Summary

CD45 expression on polymorphonuclear cells (PMNs) decreases with temperature and phagocytosis, especially in lupus patients. Anti-dsDNA antibodies enhance this clearance effect.

Background:

  • CD45, a protein tyrosine phosphatase, regulates cell functions.
  • The role of CD45 in polymorphonuclear cell (PMN)-mediated clearance of dead cells is not fully understood.

Purpose of the Study:

  • To investigate the influence of temperature and phagocytosis on CD45 expression in PMNs from lupus erythematosus (LE) patients and healthy controls.
  • To analyze the effect of anti-dsDNA antibodies on CD45 expression during phagocytosis.

Main Methods:

  • Flow cytometry was used to analyze CD45 expression on PMNs.
  • Experiments were conducted at different temperatures (4°C) and in the presence of cellular targets (nuclei, dead cells).
  • PMNs were incubated with targets pretreated with anti-dsDNA antibody-containing serum or normal serum.

  • CD45 expression was significantly down-regulated at 4°C in controls, but not in LE patients.
  • Phagocytosis led to a significant decrease in CD45 expression, more pronounced with nuclei than dead cells, and on LE PMNs compared to controls.
  • Anti-dsDNA antibodies further reduced CD45 expression, an effect amplified in LE patients.
  • Phagocytosis showed an inverse correlation with CD45 expression (P<0.0007).

Conclusions:

  • Phagocytic clearance of cellular debris by PMNs modulates effector cell function.
  • Anti-dsDNA antibodies appear to enhance the phagocytic clearance capacity of PMNs, particularly in lupus patients.
