This study used serial analysis of gene expression (SAGE) to map mouse testis gene profiles. Aging caused testicular atrophy, but gene expression remained largely stable, except for reduced protamine 2 in SAMP1 mice.

Area of Science:

  • Reproductive Biology
  • Genomics
  • Aging Research

Background:

  • The mouse testis transcriptome provides insights into male reproductive health.
  • Understanding age-related changes in gene expression is crucial for identifying reproductive decline mechanisms.

Purpose of the Study:

  • To generate a comprehensive gene expression profile of the mouse testis using SAGE.
  • To identify senescence-dependent alterations in testicular gene expression.
  • To establish a transcriptome reference for the mouse testis.

Main Methods:

  • Serial Analysis of Gene Expression (SAGE) was performed on testes from young (3-month-old) and old (29-month-old) BDF1 mice, and middle-aged (14-month-old) SAMP1 mice.
  • Quantitative analysis of SAGE tags identified differentially expressed genes.

Related Experiment Videos

  • Morphological examination assessed age-related testicular changes.
  • Main Results:

    • SAGE yielded 61,929 tags (19,323 unique), revealing high expression of genes involved in spermatogenesis, metabolism, growth, and signal transduction.
    • Aged testes showed atrophy, reduced seminiferous epithelium, and fewer spermatids/spermatocytes.
    • No significant global gene expression changes were observed in old BDF1 mice testes, but SAMP1 mice exhibited a ~50% decrease in protamine 2 gene transcription.

    Conclusions:

    • Aging induces testicular atrophy but does not grossly alter the overall gene expression profile in BDF1 mice.
    • Reduced protamine 2 expression in SAMP1 mice correlates with oligozoospermia and early reproductive decline.
    • This study provides the first quantitative mouse testis transcriptome, serving as a valuable reference.
