Combining ezetimibe and rosuvastatin significantly lowers LDL cholesterol more than rosuvastatin alone. This combination therapy for hypercholesterolemia was found to be safe and well-tolerated in patients.

Area of Science:

  • Cardiovascular Pharmacology
  • Lipid Metabolism Disorders

Background:

  • Ezetimibe is a lipid-lowering agent used with statins or alone for hypercholesterolemia.
  • Rosuvastatin is a new statin used in combination therapy.

Purpose of the Study:

  • Evaluate pharmacodynamic effects and safety of ezetimibe and rosuvastatin co-administration.
  • Examine potential pharmacokinetic interactions between ezetimibe and rosuvastatin.

Main Methods:

  • Randomized, single-blind, placebo-controlled study in hypercholesterolemic subjects.
  • Four treatment groups: rosuvastatin 10mg + ezetimibe 10mg, rosuvastatin 10mg + placebo, ezetimibe 10mg + placebo, and placebo.
  • Lipid levels assessed over 14 days; pharmacokinetic analysis conducted.

Main Results:

Related Experiment Videos

  • Co-administration of ezetimibe and rosuvastatin significantly reduced LDL-C and total cholesterol more than monotherapy.
  • Mean LDL-C reduction was -61.4% with combination therapy vs. -44.9% with rosuvastatin alone.
  • No significant pharmacokinetic interactions or adverse effects on liver/muscle function were observed.

Conclusions:

  • Ezetimibe 10 mg combined with rosuvastatin 10 mg daily provides significant incremental LDL-C reduction.
  • The combination of ezetimibe and rosuvastatin is well-tolerated in hypercholesterolemic patients.
