Summary

Yeast cytochrome c (cyt c) binds the alcohol oxidase 2 upstream activation sequence (AOX2UAS). This sequence-specific DNA-binding protein may play a regulatory role in the yeast nucleus.

Area of Science:

  • Molecular Biology
  • Yeast Genetics
  • Protein-DNA Interactions

Background:

  • The regulation of gene expression in methylotrophic yeast like Pichia pastoris is crucial for methanol metabolism.
  • Upstream activation sequences (UAS) are key regulatory elements in yeast promoters.
  • The specific proteins interacting with the AOX2UAS are not fully characterized.

Purpose of the Study:

  • To identify the protein that binds to the alcohol oxidase 2 upstream activation sequence (AOX2UAS) in Pichia pastoris.
  • To characterize the DNA-binding properties of the identified protein.
  • To investigate the potential nuclear regulatory role of this protein.

Main Methods:

  • Purification of proteins from Pichia pastoris.
  • DNA-binding assays using the AOX2UAS.

Related Experiment Videos

  • Site-directed mutagenesis of the AOX2UAS.
  • Purification and testing of cytochrome c from P. pastoris and Saccharomyces cerevisiae.
  • Main Results:

    • A protein binding to the AOX2UAS was purified and identified as cytochrome c (cyt c).
    • Cytochrome c from both Pichia pastoris and Saccharomyces cerevisiae demonstrated binding to the AOX2UAS.
    • Specific mutations within the AOX2UAS abolished cytochrome c binding, confirming sequence specificity.

    Conclusions:

    • Yeast cytochrome c is a sequence-specific DNA-binding protein.
    • Cytochrome c may function as a nuclear regulatory factor for genes containing the AOX2UAS.
    • This finding suggests a novel, non-canonical role for cytochrome c in transcriptional regulation.
