Summary
Octacalcium phosphate (OCP) combined with recombinant human bone morphogenetic protein-2 (rhBMP-2) significantly enhances bone regeneration in rat calvarium defects. This OCP/rhBMP-2 composite may allow for reduced effective doses of rhBMP-2 in bone healing applications.
Area of Science:
- Biomaterials Science
- Regenerative Medicine
- Orthopedic Research
Background:
- A bioactive and resorbable scaffold is crucial for delivering recombinant human bone morphogenetic protein-2 (rhBMP-2) to promote osteoinduction.
- Synthetic octacalcium phosphate (OCP) has demonstrated bone regeneration enhancement and resorption, being replaced by new bone.
Purpose of the Study:
- To investigate the efficacy of an OCP/rhBMP-2 composite implant in enhancing bone regeneration.
- To determine if OCP serves as an effective scaffold for rhBMP-2 delivery.
Main Methods:
- A critical-sized calvarial defect was created in rats.
- Implants consisted of OCP with either 10 microg or 1 microg of rhBMP-2, or OCP alone.
- Histomorphometrical analysis was used to quantify newly formed bone percentage (n-Bone%) at 4 and 8 weeks.
Main Results:
- OCP/rhBMP-2 implants showed significantly higher n-Bone% compared to OCP alone at both time points.
- No significant difference in n-Bone% was observed between the two rhBMP-2 dosage groups (10 microg vs. 1 microg).
Conclusions:
- OCP is an effective scaffold for rhBMP-2 delivery, enhancing bone regeneration.
- The OCP/rhBMP-2 delivery system has the potential to reduce the required effective dose of rhBMP-2 for bone healing.
