Background:

  • Pharmacologic blockade of tumor necrosis factor-alpha (TNFα) is a cornerstone therapy for autoimmune diseases like rheumatoid arthritis and Crohn's disease.
  • Agents such as etanercept and infliximab inhibit TNFα, but profound immunosuppression and risks of opportunistic infections and malignancies have been noted.
