Summary

Human hair keratin (HHK) artificial tendons promote native tendon regeneration by stimulating stem cell proliferation and differentiation. This forms the basis of in vivo tissue engineering for defect repair.

Area of Science:

  • Biomaterials Science
  • Regenerative Medicine
  • Tissue Engineering

Context:

  • Human hair keratin (HHK) artificial tendons have been utilized for over a decade in clinical applications.
  • Degradation products of HHK scaffolds stimulate tenoblast proliferation and differentiation into tenocytes.
  • Observed "in situ construction" of tissue/organ substitutes in bone, nerve, and muscle defects.

Purpose:

  • To propose a novel theoretical framework: in vivo tissue engineering.
  • To define in vivo tissue engineering as the in vivo reconstruction of defected tissues or organs.
  • To elucidate the mechanisms by which scaffold biomaterials and their degradation products facilitate tissue regeneration.

Summary:

  • Absorbable scaffold biomaterials, like HHK, activate surrounding adult stem cells for mitosis, proliferation, and differentiation.

Related Experiment Videos

  • These activated stem cells interact with the scaffold material, forming an organic complex under physiological conditions.
  • The scaffold matrix is ultimately replaced by this complex, resulting in anatomically and histologically similar native tissue.
  • Impact:

    • In vivo tissue engineering enables "in situ construction" and "in vivo cultivation" of tissue/organ substitutes.
    • This approach overcomes challenges such as immune rejection, seed cell variation, and high costs.
    • The technique offers significant clinical potential and theoretical value for regenerative medicine.
