Summary
Histone deacetylase 1 (HDAC1) mRNA levels did not differ between lung cancer and normal tissues but were higher in advanced stages. HDAC1 expression correlates with tumor progression and MTA1 in lung cancer.
Area of Science:
- Oncology
- Molecular Biology
- Gene Expression
Background:
- Histone deacetylases (HDACs) are implicated in tumorigenesis.
- The expression of HDACs in lung cancer tissues remains largely uncharacterized.
- Investigating HDAC1 and MTA1 mRNA levels provides insight into lung cancer molecular pathology.
Purpose of the Study:
- To quantify HDAC1 mRNA levels in lung cancer tissues and adjacent non-malignant tissues.
- To correlate HDAC1 mRNA expression with clinical and pathological parameters of lung cancer.
- To explore the relationship between HDAC1, MTA1 mRNA, and HDAC1 protein expression.
Main Methods:
- Quantitative analysis of HDAC1 and MTA1 mRNA using real-time reverse transcription-polymerase chain reaction (RT-PCR) in 102 lung cancer cases.
- Comparison of HDAC1/GAPDH mRNA levels between tumor and adjacent non-malignant tissues.
- Correlation analysis of HDAC1/GAPDH mRNA levels with clinicopathological data, MTA1/GAPDH mRNA, and HDAC1 protein expression via immunohistochemistry.
Main Results:
- HDAC1/GAPDH mRNA levels showed no significant difference between lung cancer and adjacent non-malignant tissues (P = 0.1827).
- HDAC1/GAPDH mRNA levels were significantly higher in advanced stages (Stage III/IV) compared to Stage I lung cancer (P = 0.0472).
- HDAC1/GAPDH mRNA levels positively correlated with MTA1/GAPDH mRNA levels (P = 0.0352) and HDAC1 protein expression (P = 0.0484).
Conclusions:
- HDAC1 gene expression may correlate with the progression of lung cancers, particularly in advanced stages.
- The correlation between HDAC1 and MTA1 suggests a potential functional relationship in lung tumorigenesis.
- Further research is warranted to elucidate the impact of HDAC1 as a molecular target in lung cancer treatment.

Histone Modification
The histone proteins have a flexible N-terminal tail extending out from the nucleosome. These histone tails are often subjected to post-translational modifications such as acetylation, methylation, phosphorylation, and ubiquitination. Particular combinations of these modifications form “histone codes” that influence the chromatin folding and tissue-specific gene expression.

lncRNA - Long Non-coding RNAs
In humans, more than 80% of the genome gets transcribed. However, only around 2% of the genome codes for proteins. The remaining part produces non-coding RNAs which includes ribosomal RNAs, transfer RNAs, telomerase RNAs, and regulatory RNAs, among other types. A large number of regulatory non-coding RNAs have been classified into two groups depending upon their length – small non-coding RNAs, such as microRNA, which are less than 200 nucleotides in length, and long non-coding RNA (lncRNA)...
