Summary

Galectin-3 is not a reliable marker for distinguishing benign from malignant thyroid lesions. While present in many thyroid cancers, it is also found in non-cancerous conditions, limiting its diagnostic utility.

  • Distinguishing between benign and malignant thyroid nodules is crucial for appropriate patient management.

Purpose of the Study:

  • To evaluate the diagnostic accuracy of galectin-3 expression in differentiating thyroid carcinomas from benign follicular lesions.
  • To assess galectin-3 as a sensitive indicator for thyroid malignancy in preoperative fine-needle aspirates (FNAs).

Main Methods:

  • Immunohistochemistry was used to assess galectin-3 protein expression in 124 formalin-fixed thyroid tissue samples.
  • Samples included papillary carcinoma, follicular carcinoma, follicular adenoma, and multinodular goitre.
  • Western blotting was performed on 24 fresh thyroid tissues to confirm antibody specificity and protein expression.

  • Galectin-3 was expressed in a high percentage of papillary (92%) and follicular (74%) carcinomas.
  • Significant expression was also observed in follicular adenomas (72%) and multinodular goitres (57%).

Conclusions:

  • Galectin-3 is not a reliable immunohistochemical marker for distinguishing benign from malignant thyroid follicular lesions.
  • The overlap in expression between malignant and benign thyroid conditions limits its utility as a standalone diagnostic marker.
