Background:

  • Constipation is a prevalent gastrointestinal condition with inadequate treatment options.
  • Tegaserod, a 5-HT4 receptor agonist, shows promise for irritable bowel syndrome with constipation.

Purpose of the Study:

  • To review efficacy and safety of tegaserod in chronic constipation patients.
  • To analyze data from two large, randomized, placebo-controlled trials.

Main Methods:

  • Two double-blind, randomized, placebo-controlled trials (E2301 and E2302) were conducted.
  • 2,612 patients received tegaserod (2 mg or 6 mg b.d.) or placebo for 12 weeks after a 2-week baseline.

Main Results:

  • Tegaserod groups showed significantly higher responder rates versus placebo in weeks 1-4 and 1-12.
  • Statistically significant improvements were observed in most secondary efficacy variables.
  • Tegaserod was safe, with no consistent differences in adverse events across groups.

Conclusions:

  • Tegaserod treatment significantly improved bowel frequency and constipation symptoms.
  • Tegaserod was well-tolerated in patients with chronic constipation.
