Donepezil significantly improved memory in multiple sclerosis (MS) patients with cognitive impairment. Further multicenter trials are recommended to confirm donepezil

Background:

  • Cognitive dysfunction is a common and disabling symptom in multiple sclerosis (MS).
  • Current treatments for MS-related cognitive impairment are limited.
  • Donepezil, an acetylcholinesterase inhibitor, is approved for Alzheimer's disease and may benefit MS patients.

Purpose of the Study:

  • To evaluate the efficacy of donepezil in improving memory and cognitive function in patients with multiple sclerosis (MS).

Main Methods:

  • A single-center, double-blind, placebo-controlled trial involving 69 MS patients with cognitive impairment.
  • Participants received either donepezil (10 mg daily) or placebo for 24 weeks.
  • Neuropsychological assessments, including the Selective Reminding Test (SRT), were performed at baseline and after treatment.

  • Donepezil treatment led to significant improvements in verbal learning and memory on the SRT compared to placebo (p=0.043).
  • Patients receiving donepezil reported memory improvement more than twice as often as placebo recipients (p=0.006).

Conclusions:
