We developed EasyTips, disposable microtips for sample preparation using photopolymerization. These tips enhance sensitivity for mass spectrometry-based proteomic analysis and phosphopeptide enrichment.

Area of Science:

  • Analytical Chemistry
  • Biochemistry
  • Materials Science

Background:

  • Effective sample preparation is crucial for sensitive mass spectrometry (MS)-based proteomic analysis.
  • Miniaturized devices for handling minute biological samples (< 10 µL) are needed for efficient analysis.
  • Current methods for sample preparation can be complex and labor-intensive.

Purpose of the Study:

  • To develop a novel, simple, and cost-effective method for fabricating disposable microtips for biological sample preparation.
  • To immobilize chromatography beads (C18 and IMAC) onto plastic pipette tips for enhanced MS analysis.
  • To demonstrate the utility of these microtips for proteomic analysis and phosphopeptide enrichment.

Main Methods:

  • Fabrication of disposable plastic microtips (

  • Immobilization of C18 reversed-phase and ion metal affinity chromatography (IMAC) beads onto polypropylene pipette tips.
  • Application of EasyTips for sample preparation in mass spectrometry (MS)-based proteomic analysis.
  • Main Results:

    • Near 100% recovery of tryptic hemoglobin digest with C18 EasyTips (25 fmol).
    • Loading capacity estimated at 0.4–2.0 µg of total proteins/peptides, sufficient for MS analysis.
    • Enhanced detection sensitivity and sequence coverage for hemoglobin digest and EGFR protein using C18 EasyTips.
    • Successful extraction and identification of phosphopeptides using IMAC EasyTips prior to MS analysis.

    Conclusions:

    • EasyTips offer a simple, cheap, and efficient method for sample preparation in MS-based proteomics.
    • The fabrication method is automatable, enabling high-throughput analysis.
    • EasyTips significantly improve detection sensitivity and facilitate phosphopeptide enrichment.
