Background:

  • The CD45 (PTPRC) 77C-->G polymorphism's role in multiple sclerosis (MS) is debated.
  • Conflicting results exist regarding its association with MS susceptibility.

Purpose of the Study:

  • To investigate the transmission of the PTPRC 77C-->G polymorphism's 'G' allele in families with MS.
  • To clarify the genetic contribution of this polymorphism to MS etiology.

Main Methods:

  • Analysis of 176 families using linkage and association statistics.
  • Utilized the TRANSMIT program to assess allele transmission patterns.

Main Results:

  • The 'G' allele was transmitted to affected offspring in 5 out of 7 families carrying the allele.
  • A statistically significant association (P=0.0342) was detected by the TRANSMIT program.

Conclusions:

  • The 77C-->G PTPRC polymorphism is present and preferentially transmitted in a small subset (<5%) of MS families.
  • Complementary analytical methods may be needed to detect this association effectively.
