[Cloning and permanent expression in eukaryotic cells of rhTpo]
1Beijing Institute of Radiation Medicine, Academy of Military Medical Sciences, Beijing.
Objective:
To obtain recombinant human thrombopoietin (rhTpo).
Methods:
rhTpo gene was cloned from fetal liver mRNA by means of RT-PCR and subcloned into permanent expressing plasmid RC/CMV in order to form a recombinant permanent expressing plasmid RC/CMV/Tpo.
Results And Conclusion:
After being transferred into CHO cells, the eukaryotic permanent expression of rhTpo, which can enhance murine megakaryocyte colony growth in vitro, was obtained.
