Summary

Calcium ions (Ca2+) promote hepatitis B virus (HBV) core assembly. The hepatitis B virus X protein (HBx) increases cytosolic Ca2+, enhancing HBV replication by boosting capsid formation.

Area of Science:

  • Virology
  • Molecular Biology
  • Cell Biology

Background:

  • Hepatitis B virus X protein (HBx) is known to influence hepatitis B virus (HBV) replication.
  • Previous studies indicated that cytosolic calcium ions (Ca2+) induced by HBx promote HBV replication.

Purpose of the Study:

  • To investigate the role of calcium ions (Ca2+) in hepatitis B virus (HBV) core assembly.
  • To elucidate the mechanism by which the hepatitis B virus X protein (HBx) influences HBV core assembly through calcium signaling.

Main Methods:

  • In vitro studies using sucrose density gradient and analytical ultracentrifuge analysis to assess HBV core assembly.
  • Transmission electron microscopy (TEM) to visualize assembled HBV core particles (capsids).
  • Ex vivo experiments using HepG2 cells treated with BAPTA-AM, cyclosporine A (CsA), and Thapsigargin (TG) to modulate intracellular calcium levels.

Related Experiment Videos

Main Results:

  • Calcium ions (Ca2+) were found to promote HBV core assembly in vitro.
  • TEM confirmed that the assembled particles were HBV capsids.
  • Inhibition of calcium signaling (BAPTA-AM, CsA) reduced HBV capsids, while calcium release (TG) increased them in HepG2 cells.
  • The hepatitis B virus X protein (HBx) was shown to increase cytosolic calcium ions, thereby promoting HBV core assembly.

Conclusions:

  • Cytosolic calcium ions (Ca2+) play a critical role in promoting hepatitis B virus (HBV) core assembly.
  • The hepatitis B virus X protein (HBx) enhances HBV replication by increasing cytosolic Ca2+, which in turn facilitates HBV capsid formation.
