Summary

Peroxynitrite, a reactive molecule formed from nitric oxide and superoxide, damages critical biomolecules. Strategies to reduce its precursors or peroxynitrite itself can mitigate drug-induced toxicity.

Area of Science:

  • Biochemistry
  • Toxicology
  • Pharmacology

Background:

  • Peroxynitrite is a potent oxidant and nitrating agent formed from nitric oxide and superoxide radicals.
  • It reacts with vital biomolecules like proteins, lipids, and DNA, altering cellular functions.
  • Excessive peroxynitrite overwhelms antioxidant defenses, leading to cell death and disrupted homeostasis.

Purpose of the Study:

  • To elucidate the role of peroxynitrite in drug-induced toxicity.
  • To explore pharmacological strategies for mitigating peroxynitrite-mediated cellular damage.

Main Methods:

  • Review of experimental evidence linking peroxynitrite to drug toxicity.
  • Analysis of mechanisms of peroxynitrite formation in drug-induced adverse effects.
  • Evaluation of potential therapeutic targets for peroxynitrite-mediated toxicity.

Main Results:

  • Peroxynitrite contributes to the toxicity of drugs such as doxorubicin, paraquat, acetaminophen, and MPTP.
  • Drug toxicity mechanisms involve enhanced peroxynitrite formation via redox cycling, NOS uncoupling, or reduced antioxidant defenses.
  • Evidence supports peroxynitrite's role in disrupting cell homeostasis and inducing cell death.

Conclusions:

  • Pharmacological interventions targeting peroxynitrite formation or scavenging are promising for attenuating drug toxicity.
  • Inhibiting precursor radical formation (e.g., using NOS inhibitors, SOD mimetics) can reduce peroxynitrite levels.
  • Directly scavenging or decomposing peroxynitrite offers another therapeutic avenue against drug-induced adverse effects.
