Summary

A novel antisense transcript, Dnm3os, was discovered within the mouse Dnm3 gene. Its expression is linked to the Twist transcription factor during development, suggesting a regulatory role for this noncoding RNA.

Area of Science:

  • Developmental Biology
  • Genetics
  • Molecular Biology

Background:

  • The mouse Dnm3 gene plays a role in development.
  • The basic helix-loop-helix transcription factor Twist is crucial for mouse development.

Purpose of the Study:

  • To identify genes regulated by the Twist transcription factor.
  • To characterize a novel antisense transcript within the Dnm3 gene.

Main Methods:

  • Gene screening
  • Transcript analysis
  • Expression profiling in wild-type and Twist-null embryos

Main Results:

  • A 6-kb antisense transcript (Dnm3os) was identified within a Dnm3 intron.
  • Dnm3os is conserved across vertebrates and does not encode a protein.

Related Experiment Videos

  • Dnm3 and Dnm3os expression overlap during embryogenesis and in adult tissues.
  • Both Dnm3 and Dnm3os are downregulated in the branchial arch of Twist-null embryos.
  • Conclusions:

    • The noncoding Dnm3os transcript is conserved and has a restricted expression pattern, suggesting a functional role in embryonic development.
    • A connection between Dnm3 regulation and the intronic antisense transcript Dnm3os is suggested.
    • Dnm3os may be regulated by the Twist transcription factor during mouse development.
