Background:

  • B cells are present in the normal human thymic medulla, concentrated around Hassall's corpuscles.
  • These B cells constitute approximately 33% of thymic medullary cells.
  • They exhibit a distinct phenotype compared to B cells found in myasthenia gravis.

Purpose of the Study:

  • To characterize human thymic B cells and their interaction with thymocytes.
  • To investigate the functional properties and potential role of these B cells in thymic selection processes.

Main Methods:

  • Immunohistochemistry and flow cytometry to identify and quantify B cells.
  • Culture of isolated thymic B cells with mitogens.
  • Electron microscopy to study B cell-thymocyte interactions.
  • Assessment of immunoglobulin expression (IgD, IgM, IgG, IgA) and CD5 marker.

Main Results:

  • Human thymic B cells express cytoplasmic IgD, IgM, and IgG, but rarely IgA, and are CD5-.
  • Isolated B cells are activated but become quiescent and die in culture without stimulation.
  • Activated B cells maintain close associations with thymocytes, including rosettes and cytoplasmic connections.
  • Two distinct thymocyte populations were observed associated with B cells via membrane or cytoplasmic contact.

Conclusions:

  • Human thymic B cells are a distinct population interacting closely with thymocytes.
  • The observed association supports the hypothesis that B cells are involved in thymic negative selection.
  • Further research is needed to fully elucidate the functional role of these B cells in T cell development.
