Area of Science:

  • Blood banking
  • Immunology
  • Diagnostic assays

Background:

  • Solid phase red cell adherence (SPRCA) assays are crucial for blood group serology.
  • Current methods often require refrigerated storage for reagent red blood cells.
  • Technological advancements enable new formats for diagnostic reagents.

Purpose of the Study:

  • To evaluate the efficacy of novel, room-temperature-stable dried reagent red blood cells for antibody screening and identification.
  • To compare the performance of dried SPRCA panels against conventional hemagglutination techniques.

Main Methods:

  • Development of immobilized and dried red cell monolayers for SPRCA.
  • Evaluation of 12-member dried RBC panels with 176 patient samples.
  • Comparison of results with established antibody specificities determined by hemagglutination.

Related Experiment Videos

Main Results:

  • Dried SPRCA panels demonstrated higher sensitivity and reduced time compared to hemagglutination.
  • Red cell antigens on dried membranes remained stable and reactive after 120 days of room temperature storage.
  • The developed reagent red cells are suitable for routine antibody screening and identification.

Conclusions:

  • Room-temperature-stable dried reagent red blood cells are a viable and improved alternative for antibody screening and identification.
  • This technology enhances the efficiency and accuracy of pre-transfusion testing.
  • The stability of dried RBCs simplifies logistics and expands the usability of SPRCA assays.
