Summary

Insulin-like factor 3 (INSL3) is crucial for testicular descent in males. This peptide hormone also marks the mature phenotype of Leydig cells and ovarian theca interna cells.

Area of Science:

  • Endocrinology
  • Reproductive Biology
  • Molecular Endocrinology

Background:

  • Insulin-like factor 3 (INSL3), also known as relaxin-like factor (RLF), is a peptide hormone produced by testicular Leydig cells in men and ovarian theca interna cells in women.
  • INSL3 is structurally similar to insulin and relaxin but signals through a distinct G-protein-coupled receptor, LGR8.
  • INSL3 plays a vital role in the transabdominal phase of testicular descent, with its absence causing cryptorchidism.

Purpose of the Study:

  • To characterize Insulin-like factor 3 (INSL3) as a marker for differentiated Leydig and theca interna cells.
  • To develop antibodies for INSL3 detection via immunohistochemistry and immunoassays.

Main Methods:

  • Development of species-specific antibodies for INSL3.

Related Experiment Videos

  • Application of antibodies in immunohistochemistry and immunoassays.
  • Analysis of INSL3 expression in mature Leydig cells and ovarian theca interna cells.
  • Main Results:

    • INSL3 is constitutively expressed in mature Leydig cells once differentiation is achieved.
    • INSL3 serves as an indicator of differentiation status for both Leydig cells and ovarian theca interna cells.
    • Antibodies developed are suitable for immunohistochemistry and immunoassays, facilitating INSL3 detection.

    Conclusions:

    • INSL3 is a key marker for the mature phenotype of testicular Leydig cells.
    • INSL3 is also a reliable indicator of differentiation status in ovarian theca interna cells.
    • The developed INSL3 antibodies are valuable tools for studying this hormone and related cellular differentiation.
