Summary

The fission yeast nucleus, positioned by microtubules, determines the cell division plane. Displacing the nucleus before mitosis causes asymmetric division, revealing its critical role in cell division accuracy.

Area of Science:

  • Cell biology
  • Microbiology
  • Genetics

Background:

  • Cell division plane positioning is crucial for cell shape, size, and tissue organization.
  • In Schizosaccharomyces pombe, the nucleus is centrally located and division occurs at the cell center.
  • Microtubules (MTs) are essential for maintaining the nucleus's central position.

Purpose of the Study:

  • To investigate the role of microtubules in nuclear positioning.
  • To determine the nucleus's role in positioning the division plane.
  • To understand how nuclear displacement affects cell division symmetry.

Main Methods:

  • Utilizing optical tweezers to experimentally displace the nucleus in Schizosaccharomyces pombe.
  • Observing the nucleus's return to the cell center via MTs.

Related Experiment Videos

  • Analyzing the impact of nuclear displacement at different cell cycle stages on division plane positioning.
  • Main Results:

    • Displaced nuclei were repositioned to the cell center by MTs pushing against cell tips.
    • Nuclear displacement during interphase or early prophase led to asymmetric cell division.
    • Displacement during prometaphase resulted in symmetric division, similar to control cells.

    Conclusions:

    • The predividing nucleus specifies the cell division plane.
    • Nuclear positioning by MTs is critical for accurate, symmetric cell division in fission yeast.
    • Establishing the division plane early in mitosis is an optimal strategy for symmetric division.
