Plasmacytoid dendritic cells (pDCs) are crucial for generating antiviral cytotoxic T lymphocytes (CTLs) during herpes simplex virus (HSV) infection. These pDCs collaborate with lymph node dendritic cells (LNDCs) to effectively clear the virus.

Background:

Antiviral immunity relies on T cells, particularly cytotoxic T lymphocytes (CTLs), for pathogen clearance.
Dendritic cells (DCs) orchestrate adaptive immune responses, with plasmacytoid DCs (pDCs) known for interferon-alpha production.

Purpose of the Study:

To elucidate the role of lymph node-recruited pDCs in antiviral T cell immunity against herpes simplex virus (HSV).
To investigate how pDCs collaborate with lymph node dendritic cells (LNDCs) in initiating antiviral CTL responses.

Main Methods:

Utilized a murine model of cutaneous HSV infection.
Employed in vivo pDC depletion and transfer of pDC precursors.
Conducted in vitro co-culture experiments with LNDCs and T cells.
