    Summary

    Mitochondria from rat hearts are highly sensitive to calcium-induced permeability transition pore opening shortly after isolation. This sensitivity decreases over time, correlating with changes in the calcium uniporter kinetics.

    • Incubation for 2-4 hours post-isolation leads to a decrease in mitochondrial sensitivity to PTP opening.
    • This decreased sensitivity correlates with an increase in apparent K(Ca) values (up to 0.4 microM).

    Conclusions:

    • Mitochondrial sensitivity to Ca2+-induced PTP opening is highest shortly after isolation and diminishes over time.
    • Changes in the kinetic parameters of the Ca2+-uniporter are associated with altered mitochondrial sensitivity to calcium-induced permeability transition.
