Summary

This study investigated how indole acetic acid (IAA), epidermal growth factor (EGF), and follicle-stimulating hormone (FSH) affect sheep ovarian follicle growth and viability in vitro. Combining IAA with EGF or EGF with FSH best supported activated primordial follicle health during culture.

Background:

  • Mechanisms regulating primordial follicle exit from the non-growing pool are poorly understood.
  • Ovarian follicle activation is crucial for fertility and assisted reproductive technologies.

Purpose of the Study:

  • To evaluate the effects of indole acetic acid (IAA), epidermal growth factor (EGF), and follicle-stimulating hormone (FSH) on ovine preantral follicle growth and viability in vitro.
  • To determine optimal supplement combinations for maintaining follicle health during in vitro culture.

Main Methods:

  • Ovine ovarian cortex fragments were cultured in vitro for 2 or 6 days.
  • Culture media were supplemented with various combinations of IAA, EGF, and FSH.
  • Follicle classification (primordial vs. developing) and viability assessments were performed via histological examination.

Main Results:

  • All tested media supplements increased the percentage of developing follicles and decreased primordial follicles compared to controls.
  • Culture for 6 days reduced overall follicle viability, except in media supplemented with IAA+EGF or EGF+FSH.

Conclusions:

  • These findings provide insights into optimizing conditions for ovarian tissue culture and potential fertility preservation strategies.
