Background:

Purpose of the Study:

  • To investigate the androgen regulation of CD-9 protein.
  • To analyze the correlation between CD-9 expression and prostate cancer malignancy.

  • Microarray analysis of LNCaP and LNCaP-r cell lines.
  • Western blot, RT-PCR, and immunohistochemistry on cell lines and 88 human prostate cancer tissues.
  • Analysis of CD-9 expression in response to synthetic androgen R1881 and in different Gleason grades.

Main Results:

  • CD-9 expression was induced by R1881 in LNCaP cells.
  • CD-9 was localized in luminal and basal epithelial cells of non-malignant prostate tissue.
  • CD-9 staining intensity varied in tumor areas and was not related to Gleason grade; it was absent in tissues from patients under androgen ablation therapy.

Conclusions:

  • CD-9 is an androgen-regulated protein present in human prostate specimens.
  • Variable CD-9 expression and localization suggest a potential role in prostate cancer development and progression.
