Summary

Pharmacological kindling from lidocaine hydrochloride (HCl) in mice appears to depend more on the total number of injections than their frequency. Doses around 40 mg/kg approached the threshold for inducing kindling.

Background:

  • Lidocaine hydrochloride (HCl) is known to induce pharmacological kindling in rats when administered frequently.
  • Understanding the dose-response and frequency-dependence of this effect is crucial for preclinical research.

Purpose of the Study:

  • To determine the approximate threshold dose for lidocaine-induced pharmacological kindling in mice.
  • To investigate whether a reduced injection frequency impacts the development of kindling.

Main Methods:

  • Mice received intraperitoneal injections of lidocaine HCl at doses ranging from 30 to 50 mg/kg.
  • Two injection schedules were employed: 5 times weekly for 4 weeks, or 2 times weekly for 10 weeks.
  • Kindling was defined as observing convulsions in 5 consecutive injections.

Main Results:

  • The estimated threshold dose for kindling was approximately 40 mg/kg, with 2/8 mice kindled at this dose and 8/8 at 50 mg/kg when injected 5 times weekly.
  • When administered at 50 mg/kg, 80% of mice were kindled by the 15th injection, regardless of whether injections were given 5 times or twice weekly.

Conclusions:
