Summary
New Ruthenium(III) complexes with maltolato, ethylmaltolato, and pyridonato ligands were synthesized and characterized. These novel compounds, particularly Ru(III) ethylmaltolato, show promising anti-proliferative activity against human breast cancer cells.
Area of Science:
- Inorganic Chemistry
- Medicinal Chemistry
- Materials Science
Background:
- Ruthenium complexes are investigated for their potential therapeutic applications, including anticancer properties.
- Water-soluble coordination compounds offer advantages for biological studies and drug delivery.
Purpose of the Study:
- To synthesize and characterize novel water-soluble Ruthenium(III) complexes with maltolato, ethylmaltolato, and pyridonato ligands.
- To evaluate the anti-proliferative activity of these synthesized complexes against human breast cancer cell lines.
Main Methods:
- Synthesis of Ru(III) complexes: Ru(ma)3 (1a), Ru(etma)3 (1b), and Ru(pyd)3 (2a).
- Characterization techniques: Elemental analysis, NMR (1H, 13C, COSY, HMQC), IR spectroscopy, Mass Spectrometry (MS), solution conductivity, cyclic voltammetry, and X-ray crystallography.
- Anti-proliferative assays: Testing against the MDA-MB-435S human breast cancer cell line.
Main Results:
- The Ru(III) complexes were successfully synthesized and characterized, confirming their structures.
- X-ray crystallography of Ru(ma)3 revealed a mer configuration.
- Complexes 1a and 1b predominantly exist as mer isomers in aqueous solution, while 2a is a mixture of mer and fac isomers.
- Complexes 1a and 1b exhibited anti-proliferative activity with IC50 values of 140 µM and 90 µM, respectively, against MDA-MB-435S cells.
Conclusions:
- The synthesized water-soluble Ru(III) complexes are structurally diverse and exhibit isomerism in solution.
- Ru(III) ethylmaltolato complex demonstrates significant anti-proliferative effects on human breast cancer cells, warranting further investigation for potential therapeutic development.

Chirality at Nitrogen, Phosphorus, and Sulfur
Chirality is most prevalent in carbon-based tetrahedral compounds, but this important facet of molecular symmetry extends to sp3-hybridized nitrogen, phosphorus and sulfur centers, including trivalent molecules with lone pairs. Here, the lone pair behaves as a functional group in addition to the other three substituents to form an analogous tetrahedral center that can be chiral.
A consequence of chirality is the need for enantiomeric resolution. While this is theoretically possible for all...

¹H NMR: Complex Splitting
A proton M that is coupled to a proton X results in doublet signals for M. However, NMR-active nuclei can be simultaneously coupled to more than one nonequivalent nucleus. When M is coupled to a second proton A, such as in styrene oxide, each peak in the doublet is split into another doublet.
Splitting diagrams or splitting tree diagrams are routinely used to depict such complex couplings. While drawing splitting diagrams, the splitting with the larger coupling constant is usually applied first.
