A new software tool analyzes photobleaching fluorescence resonance energy transfer (pbFRET) data, enabling pixel-by-pixel calculation of energy transfer values. This facilitates proximity measurements of labeled macromolecules in biological samples.

Area of Science:

  • Spectroscopy and microscopy
  • Biophysics
  • Molecular biology

Background:

  • Photobleaching fluorescence resonance energy transfer (pbFRET) is a microscopy technique for measuring macromolecule proximity.
  • pbFRET analysis requires pixel-by-pixel calculation of bleaching time constants from image series.
  • Existing software lacks the capability to process the extensive data required for pbFRET.

Purpose of the Study:

  • To develop and introduce new evaluation software for analyzing pbFRET data.
  • To address the limitations of existing software in processing pbFRET image series.
  • To enable efficient and comprehensive analysis of donor photobleaching data.

Main Methods:

  • Developed new evaluation software for Windows using National Instrument LabVIEW 6.1.

Related Experiment Videos

  • Integrated a mathematical virtual instrument package, including the Levenberg-Marquardt routine.
  • Applied the software to determine FRET efficiency in major histocompatibility complex (MHC) glycoproteins in human B lymphoma cells.
  • Main Results:

    • The software calculates bleaching time constants on a pixel-by-pixel basis.
    • Results can be visualized as color-coded maps or histograms from raw image data.
    • Demonstrated successful application in measuring FRET efficiency for MHC molecules.

    Conclusions:

    • Introduced a new version of pbFRET analysis and data processing software.
    • The software generates a complete analysis pattern for donor photobleaching image series.
    • This tool enhances the capability to study macromolecule interactions using pbFRET.
