Objective:
The purpose of this study was to invest the effect of epidermal growth factor receptor monoclonal antibody MAb225 on radiosensitivity of tongue squamous cell carcinoma cell Tca8113.
Methods:
Tca8113 cells were treated with different concentrations of MAb225. Radiation dose survival curve was generated from clonogenic survival assay. SERPD0 and survival faction (SF2) was analysed by single hit multi-target (SHMT) radiobiological model using RADMEDIC software. Nude mice with Tca8113 tumor xenografts were treated with MAb225, radiation treatment or both of them. Tumor responses were assessed by tumor growth delay, and t test was used for statistical analysis.
Results:
MAb225 enhanced the radiosensitivity of Tca8113 cells. SERD0 of Tca8113 cells treated with 0.5 mg/L MAb225 was 1.23. The survival fraction of cells treated with MAb225 was significantly decreased. Tumor radioresponse could be enhanced by MAb225 (P < 0.05).
Conclusion:
MAb225 could enhance radiosensitivity of tongue squamous cell carcinoma cell.
Insights
Epidermal growth factor receptor monoclonal antibody MAb225 increases the effectiveness of radiation therapy for tongue squamous cell carcinoma. This antibody enhances radiosensitivity in Tca8113 cells and improves tumor response to radiation.
Area of Science:
- Oncology
- Molecular Biology
- Radiotherapy
Background:
- Tongue squamous cell carcinoma (TSCC) is a common head and neck cancer.
- Improving the efficacy of radiotherapy is crucial for better patient outcomes.
- Targeting specific cellular pathways, like EGFR, offers potential therapeutic strategies.
Purpose of the Study:
- To investigate the impact of epidermal growth factor receptor (EGFR) monoclonal antibody MAb225 on the radiosensitivity of Tca8113, a tongue squamous cell carcinoma cell line.
- To evaluate the combined effect of MAb225 and radiation on tumor response in vivo.
Main Methods:
- Tca8113 cells were treated with varying concentrations of MAb225.
- Clonogenic survival assays determined radiation dose survival curves.
- The single-hit multi-target (SHMT) radiobiological model was used to analyze sensitization enhancement ratio (SERD0) and survival fraction (SF2).
- Nude mice bearing Tca8113 tumor xenografts were treated with MAb225, radiation, or both, with tumor growth delay assessed.
Main Results:
- MAb225 significantly enhanced the radiosensitivity of Tca8113 cells.
- Treatment with 0.5 mg/L MAb225 resulted in a SERD0 of 1.23.
- The survival fraction of Tca8113 cells post-irradiation was markedly reduced in the presence of MAb225.
- MAb225 demonstrated a statistically significant enhancement of tumor radioresponse (P < 0.05).
Conclusions:
- MAb225 effectively enhances the radiosensitivity of tongue squamous cell carcinoma cells.
- The combination of MAb225 and radiation therapy shows promise for treating TSCC.
