Background:

  • Three-dimensional (3-D) reconstruction is vital for studying biological structures in embryology and developmental biology.
  • Specimen preparation for 3-D reconstruction often presents significant challenges, including mutual displacement of structures in serial sections.

Purpose of the Study:

  • To evaluate the utility of non-deparaffinized tissue sections for 3-D reconstruction.
  • To assess if retaining paraffin in sections minimizes structural displacement and facilitates reconstruction.
  • To explore the compatibility of this method with immunohistochemistry.

  • Chicken embryos were fixed, stained with eosin, and embedded in paraffin.
  • Approximately 30-µm-thick non-deparaffinized serial sections were prepared.
