Clinical doses of diazepam (DZ) did not cause tolerance in the vestibulo-ocular reflex (VOR) as measured by the sinuosidal harmonic acceleration (SHA) rotational test. This suggests patients may not need to stop DZ before SHA testing.

Background:

  • Repeated diazepam (DZ) administration is known to cause tolerance in memory, sedation, and psychomotor tasks.
  • The effect of DZ on the vestibulo-ocular reflex (VOR) has not been previously established.

Purpose of the Study:

  • To determine if clinical doses of diazepam (10 mg/d for 14 days) induce tolerance in the VOR using the sinuosidal harmonic acceleration (SHA) rotational test.

Main Methods:

  • A double-blinded, repeated-measures study involving 30 healthy males aged 20-36.
  • Subjects were randomized to placebo or DZ groups, undergoing 6 SHA rotational sessions over 2 weeks.
  • Analysis utilized a 1-way ANOVA to compare percent change from baseline between groups.

Main Results:

  • Significant differences (p < .05) in VOR gain and phase were observed at lower frequencies (0.01–0.08 Hz) between treatment groups.
