Summary

Prokineticin 2 (PK2) is a key molecule in the suprachiasmatic nucleus (SCN) that regulates circadian rhythms and sleep-wake cycles. Its molecular rhythm, influenced by light and internal loops, impacts SCN targets controlling behavior.

Area of Science:

  • Neuroscience
  • Chronobiology
  • Molecular Biology

Background:

  • Circadian timing, regulated by the suprachiasmatic nucleus (SCN), is crucial for sleep.
  • Interactions between circadian and homeostatic sleep processes are vital.
  • Prokineticins are newly identified regulatory proteins.

Purpose of the Study:

  • To summarize the biological functions of prokineticins.
  • To focus on the circadian role of prokineticin 2 (PK2).
  • To explore PK2's potential involvement in sleep-wake regulation.

Main Methods:

  • Review of animal lesion and genetic studies.
  • Analysis of prokineticin 2 (PK2) molecular rhythm in the SCN.
  • Examination of PK2 receptor expression in SCN output targets.

Main Results:

  • PK2 is a candidate SCN output molecule regulating circadian locomotor behavior.
  • PK2's SCN rhythm is controlled by transcriptional/translational loops and light.
  • PK2 receptor is expressed in SCN targets controlling sleep-wake behavior.

Conclusions:

  • PK2 plays a significant role in circadian rhythm and sleep-wake regulation.
  • The depolarizing effect of PK2 on its receptor-expressing neurons is a potential regulatory mechanism.
  • PK2 is a critical link between the SCN and behavioral outputs.
