Summary

Grey matter demyelination in multiple sclerosis (MS) is widespread, causing significant neuronal loss. This grey matter damage may explain disability and cognitive issues in MS patients.

Background:

  • Multiple sclerosis (MS) is a chronic inflammatory disease of the central nervous system.
  • Grey matter lesions, neuronal loss, and synaptic loss are implicated in MS pathogenesis and clinical symptoms.

Purpose of the Study:

  • To investigate the extent and distribution of grey matter demyelinating lesions in MS.
  • To assess associated neuronal and synaptic loss within these lesions.
  • To correlate grey matter pathology with clinical outcomes in MS.

Main Methods:

  • Analysis of whole coronal brain sections from 6 MS and 6 control cases.
  • Immunohistochemistry for myelin, neuronal, synaptic, and apoptotic markers.
  • Quantification of neuronal density and synaptophysin staining in cortical lesions versus normal cortex.
