Loss of heterozygosity (LOH) of the retinoblastoma gene (Rb) is common in high-grade chondrosarcomas, particularly dedifferentiated types. Rb-LOH correlates with absent Rb protein expression, but does not predict progression in low-grade tumors.

Area of Science:

  • Oncology
  • Molecular Biology
  • Genetics

Background:

  • Loss of function of the human retinoblastoma gene (Rb) is a frequent genetic abnormality in human malignancies.
  • Rb gene dysfunction disrupts cell cycle control, leading to abnormal proliferation and differentiation.

Purpose of the Study:

  • To investigate the loss of heterozygosity (LOH) of the Rb gene in various cartilaginous tumors.
  • To correlate Rb-LOH status with Rb protein expression via immunohistochemistry.

Main Methods:

  • Polymerase chain reaction (PCR) was used to detect Rb-LOH in 31 formalin-fixed, paraffin-embedded cartilaginous tumors.
  • Tumors included dedifferentiated chondrosarcoma, conventional chondrosarcoma (grades 1-3), enchondromas, and chondroblastomas.
  • Microdissection separated components of dedifferentiated chondrosarcoma; Rb protein expression was assessed by immunohistochemistry.

Related Experiment Videos

Main Results:

  • Rb-LOH was detected in one grade 3 chondrosarcoma and in the anaplastic component of 7 out of 8 dedifferentiated chondrosarcomas (89%).
  • All tumors with Rb-LOH showed negative Rb immunohistostaining.
  • Benign tumors and low-grade chondrosarcomas were negative for Rb-LOH and positive for Rb protein expression.

Conclusions:

  • Rb-LOH predominantly occurs in high-grade chondrosarcomas, especially dedifferentiated types.
  • Rb-LOH is associated with loss of Rb protein expression in high-grade tumors.
  • Rb-LOH is not a reliable marker for predicting progression or recurrence in low-grade cartilaginous tumors.
