Activins, TGF-beta family proteins, regulate FSH secretion and cell functions. This study used DNA microarrays to identify genes regulated by activin and GnRH, revealing distinct gene expression patterns and potential signaling pathways.
Area of Science:
- Endocrinology
- Molecular Biology
- Cell Signaling
Background:
- Activins, part of the TGF-beta superfamily, are widely expressed and influence FSH secretion, development, cell growth, apoptosis, and inflammation.
- The precise molecular mechanisms and downstream signaling pathways of activin's diverse functions are not fully understood.
Purpose of the Study:
- To identify genes regulated by activin, alone or with gonadotropin-releasing hormone (GnRH), in a murine gonadotrope cell line (LbetaT2).
- To elucidate the downstream signaling pathways utilized by activin.
Main Methods:
- Utilized DNA microarray assays (Affymetrix Mu74av2 Gene Chip) to screen gene expression in LbetaT2 cells.
- Treated cells with activin A, GnRH agonist (GnRHA), or both, and analyzed gene expression changes.
- Confirmed microarray findings for selected genes using real-time quantitative PCR and RT/PCR.
Main Results:
- Identified 268 transcripts significantly up- or down-regulated (≥2-fold) by activin, GnRHA, or their combination.
- Observed distinct gene expression profiles for each treatment, suggesting interplay between activin and GnRH signaling.
- Gene expression alterations ranged from 15 to 67-fold.
Conclusions:
- Activin modulates the expression of a wide array of genes in LbetaT2 cells.
- The findings suggest that activin mediates its diverse biological effects through multiple signaling pathways.
- Interactions between activin and GnRH signaling pathways were indicated.
