    Oncornaviruses possess phosphoproteins as structural components. A common 12,000 mol wt phosphoprotein was identified across multiple oncornavirus strains, with other specific phosphoproteins found in certain viruses.

    Background:

    • Oncornaviruses are RNA viruses known to possess virion-associated protein kinase activity.
    • Virion structural components of oncornaviruses have been investigated for their protein composition.

    Purpose of the Study:

    • To identify and characterize phosphoproteins present as structural components within various oncornavirus strains.
    • To determine the molecular weights and phosphoamino acid composition of these viral phosphoproteins.

    Main Methods:

    • Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) was used to determine molecular weights.
    • Amino acid analysis was performed to identify phosphoamino acid residues.

    Main Results:

    • A major phosphoprotein of approximately 12,000 mol wt was common to mouse and feline leukemia oncornaviruses.
    • Specific strains exhibited additional phosphoproteins: Kirsten murine sarcoma virus (10,000 mol wt), mouse erythroblastosis virus (74,000 mol wt glycoprotein), and RD-114 virus (16,000 mol wt).
    • Phosphoserine was identified in the 12,000 mol wt polypeptide of mouse erythroblastosis virus, and phosphothreonine in the 16,000 mol wt polypeptide of RD-114 virus.

    Conclusions:

    • Oncornaviruses incorporate distinct phosphoproteins as integral structural elements.
    • The identified phosphoproteins vary in molecular weight and phosphoamino acid content across different oncornavirus species.
