Background:

  • Nonrandom translocations involving chromosome 18 band q21 can deregulate the bcl-2 gene, potentially leading to neoplastic transformation in human lymphomas.
  • The expression patterns of bcl-2 in hematopoietic cells remain incompletely understood.

Purpose of the Study:

  • To investigate the expression levels of bcl-2 messenger RNA (mRNA) in various myeloid and lymphoid cell malignancies.
  • To determine if bcl-2 expression is associated with specific hematologic cancers, particularly those lacking the t(14;18) translocation.

Main Methods:

  • Molecular genetic analysis was employed to assess bcl-2 gene rearrangement.
  • Messenger RNA (mRNA) levels were measured in a diverse cohort of myeloid and lymphoid cell malignancies.

Main Results:

  • Bcl-2 gene rearrangement was detected in only 2 out of 77 patients (hairy cell leukemia and diffuse small cleaved cell lymphoma).
  • While some myeloid leukemias showed high bcl-2 levels, expression is predominantly observed in lymphoid malignancies.
  • Peak bcl-2 mRNA expression in fresh specimens was identified in differentiated B lymphocytes, characteristic of chronic lymphocytic leukemia.

Conclusions:

  • Bcl-2 expression is characteristic of certain lymphoid malignancies, even without the t(14;18) translocation.
  • The bcl-2 gene product may play a role in prolonging cell survival, contributing to the biology of disorders like chronic lymphocytic leukemia.
