Background:

  • Leishmania spp. promastigotes show varying sensitivity to complement-mediated lysis (CML) during different growth phases and culture conditions.
  • Leishmania chagasi exhibits CML resistance in stationary phase cultures from infected animals, but becomes constitutively sensitive after serial passaging.

Purpose of the Study:

  • To identify novel genetic factors conferring complement resistance in Leishmania chagasi.
  • To understand the molecular mechanisms behind Leishmania's survival against host immune defenses.

  • A genetic screen using a cosmid library was performed on a serially passaged, CML-sensitive L. chagasi promastigote line.
  • Transfection of promastigotes with the library and selection for CML resistance using human serum.

    • The identified genes, homologous to those on L. major chromosome 36, are potential targets for understanding and combating Leishmania infections.
