Seven diterpenoids were isolated from Nuxia sphaerocephala leaves. Two compounds, 3-oxolupenal and 3beta-hydroxy-lupenal, demonstrated significant anti-malarial activity against Plasmodium falciparum in vitro.
Area of Science:
- Natural Products Chemistry
- Medicinal Chemistry
- Pharmacognosy
Background:
- Nuxia sphaerocephala is a plant species with potential medicinal properties.
- Diterpenoids and lupane derivatives are classes of natural compounds known for diverse biological activities.
Purpose of the Study:
- To isolate and characterize chemical constituents from the leaves of Nuxia sphaerocephala.
- To evaluate the anti-malarial activity of the isolated compounds against Plasmodium falciparum.
Main Methods:
- Extraction of plant material using ethyl acetate.
- Isolation and purification of compounds using chromatographic techniques.
- Structure elucidation using spectroscopic methods (NMR, MS) and chemical procedures (PGME amide).
- In vitro anti-malarial activity assay against Plasmodium falciparum.
Main Results:
- Seven new diterpenoids (four clerodanes and three labdanes) were isolated, along with 17 known compounds.
- Compounds 3-oxolupenal and 3beta-hydroxy-lupenal exhibited potent inhibitory activity against Plasmodium falciparum, with IC50 values ranging from 1.55 to 4.67 microg/ml.
- The structures and relative stereochemistry of the new compounds were determined, and the absolute configuration at C-13 for two compounds was established.
Conclusions:
- Nuxia sphaerocephala leaves are a source of novel diterpenoids.
- 3-Oxolupenal and 3beta-hydroxy-lupenal represent promising lead compounds for the development of new anti-malarial drugs.
