Summary

Reduced expression of caspases 8 and 10 may play a role in trophoblast tumorigenesis. This study investigated caspase 8 and 10 in gestational trophoblastic diseases, finding lower levels in choriocarcinoma.

Placental trophoblast exhibits pseudomalignant characteristics.
The pathogenesis of gestational trophoblastic diseases requires further elucidation.

Purpose of the Study:

To investigate the role of caspases 8 and 10 in trophoblast tumorigenesis.
To analyze differential gene expression of caspases in gestational trophoblastic diseases.

Differential gene expression analysis using cDNA array hybridization.
Quantitative real-time polymerase chain reaction and immunohistochemistry for validation.
Analysis of caspase 8 and caspase 10 expression in choriocarcinoma cell lines and normal placenta.

Caspase 10 was downregulated in choriocarcinoma cell lines compared to normal placenta.
Significantly lower caspase 10 mRNA levels were observed in hydatidiform mole and choriocarcinoma.
Protein expression of caspase 8 and 10 was reduced in choriocarcinomas, correlating with apoptotic indices.

Conclusions:

Downregulation of caspases 8 and 10 is suggested to contribute to choriocarcinoma pathogenesis.
These findings highlight a potential mechanism in the development of gestational trophoblastic diseases.
